BioNTech halts Phase 2 cancer vaccine trial

BioNTech SE and Genentech (Roche) have terminated the Phase 2 trial of autogene cevumeran, an individualized mRNA cancer immunotherapy, in ctDNA-positive, surgically resected high-risk Stage II/III colorectal cancer as an adjuvant monotherapy. The decision followed a recommendation from an independent Data Safety Monitoring Board which found a numerical imbalance in overall survival between treatment arms and judged that continuing the trial was unlikely to change the outcome. BioNTech said it would analyze the data to refine patient selection and inform future mRNA oncology strategies, while noting that a separate Phase 2 trial in pancreatic cancer in combination with checkpoint inhibition and chemotherapy remains ongoing.
